The Research Group 

The project will be pursued in a lab with an excellent track record in diabetes research (headed by Nils Wierup) https://www.ludc.lu.se/nils-wierup-pi. The Wierup lab is part of the Lund University Diabetes Centre  http://www.ludc.med.lu.se/. Lund University Diabetes Centre is one of the strongest centers for diabetes research in the World.

The research Group is situated at Clinical Research Centre at the Scania University Hospital in Malmö in southern Sweden.

We are hereby adverstising for the position of a post doctoral fellow within diabetes research. The position is full time for two years according to central agreement for temporary position as Post Doctor. Desired start date is May 1st 2020 or according to agreement. 

Project Description 

Cell type-specific impact of T2D on the transcriptome will be assessed with 10X and SmartSeq2 single cell RNA sequencing in islets from T2D patients and controls using established pipelines. Cell type-specific Gene-regulatory networks (GRNs) will be computed using a novel algorithm. GRNs representing key components of beta cell pathology have been identified (using the transcriptional profiles of 5000 cells) and will now be subjected to functional validation.  Gene expression profile will be related to single cell functional properties, and potential subpopulations of alpha- and beta cells will be assessed using Patch-seq. GRNs will be functionally validated using CROP-seq and whole genome CRISPR-screens. Selected hub genes in the GRNs will be functionally validated using CRISPR-Cas9 gene targeting and state of the art techniques for assessment of beta cell function.

Work Duties 

The project will be pursued at Lund University Diabetes Centre, one of the strongest centers for T2D research in the world. A wide group of collaborators will provide expertise, including bioinformatics and development of novel algorithms.

  • CROP-seq and CRISPR-screens will be performed in collaboration with SciLifeLab, Karolinska Institutet, Stockholm.
  • The position will include a high degree of molecular biology wet lab work, patch clamp experiments as well as bioinformatic analysis and interpretation of data. 
  • As a Post Doctor you will be responsible for planning and running all experiments and finalizing scientific reports with a high degree of independence, but with close supervision by the group leaders and aid from our mentoring program.  
  • Other work duties within the research group can be performed as well. 

Qualification requirements 

A post doctor is an employee who has doctoral degree, or a foreign degree that is assessed to equal a doctoral degree. The applicant needs to have a PhD received maximum 3 years before the deadline of this application. Special reasons may apply that change this time. For employment as a Post Doctor following criteria will be applied: 

  • Good ability to develop and perfom research with good quality and educational skill. 

A Post Doctor position should foremost consider scientific ability. Other assessments will be done according to the following requirements for the position: 

  • Applicants should have a Ph.D. in a relevant area.
  • The project needs a highly motivated person with documented excellence in molecular- or computational biology.
  •  Good communication skills in the English language (written and spoken) is a prerequisite.

Merits: 

  • A strong background in the diabetes field.
  • Documented expertise in CRISPR-Cas9 technology, single cell RNAseq or patch clamp.
