Working task

The Ph D student will study protein-protein, protein-ligand interactions and protein self-assembly reactions with an aim to reach mechanistic insights into these processes. Problems that will be addressed include oligomer formation during amyloid fibrils formation reactions, monomer- and oligomer-interaction with amyloid fibrils, and the effect of inhibitors on oligomer generation. The Ph D student will develop analytical tools for understanding protein-protein interactions and protein self-assembly reactions. This includes developments of methods that use a combination of biochemical reactions, biophysical techniques and building/production of physical tools/equipment. The position will include development of tools for photo-induced crosslinking of unmodified proteins as well as tools and reaction conditions for phage retrieval from microscope slides for DNA sequencing to enable the development of massive parallel phage display.

Qualifications

The candidate shall have a master's degree in molecular protein science, chemical engineering, biotechnology, nanotechnology or equivalent. The candidate shall have work experience of performing and optimizing Photo Induced Crosslinking of Unmodified Proteins (PICUP) or equivalent and have practical experience of designing new laboratory instruments and components for 3D printing. Work experience of purification and biophysical studies of fibril-forming proteins are a merit. Completed courses in physical chemistry at the master's level are meriting.

The candidate should be well acquainted with:

Photo Induced Crosslinking of Unmodified Proteins (PICUP) or equivalent.

Molecular biological techniques such as DNA amplification, DNA electrophoresis, DNA digestion, ligation and insertion of plasmid, and modification of DNA by mutation using PCR techniques.

Techniques for expressing and purifying proteins such as protein expression in bacterial expression systems, purification by chromatographic techniques and protein electrophoresis.

Membrane system techniques such as preparation of liposomes by extrusion and analysis of protein-membrane interactions.

Biophysical techniques such as fluorescence spectroscopy, isothermal titer calorimetry (ITC) and kinetic measurements using fluorescence.

Basic NMR spectroscopy.


Eligibility
Students with basic eligibility for third-cycle studies are those who- have completed a second-cycle degree- have completed courses of at least 240 credits, of which at least 60 credits are from second-cycle courses, or- have acquired largely equivalent knowledge in some other way, in Sweden or abroad.

The employment of doctoral students is regulated in the Swedish Code of Statues 1998: 80. Only those who are or have been admitted to PhD-studies may be appointed to doctoral studentships. When an appointment to a doctoral studentship is made, the ability of the student to benefit from PhD-studies shall primarily be taken into account. In addition to devoting themselves to their studies, those appointed to doctoral studentships may be required to work with educational tasks, research and administration, in accordance with specific regulations in the ordinance.
